序列测定系统包括流体装置和光学装置，反应装置包括第一单元和第二单元，定义序列测定反应包含的一种重复执行单位为第二生化反应-第一生化反应-拍照，方法包括在完成初始步骤之后，使得当利用流体装置使第一单元和第二单元中的一个上的样品进行第二生化反应和第一生化反应的同时，利用光学装置对另一个单元上的样品进行拍照。初始步骤包括：a）利用流体装置使第一单元和第二单元中的一个进行样品的第一生化反应，b）利用光学装置对进行第一生化反应后的单元的样品的进行拍照，c）利用流体装置使第一单元和第二单元的另一个进行样品的第一生化反应。上述方法可提高序列测定效率。
“The present disclosure discloses a method for controlling a base sequence determination, a base sequence determination system, and a control device. 本专利公开了一种全内反射荧光成像系统及测序装置，全内反射荧光成像系统包括第一成像系统和第二成像系统，所述第一成像系统包括N条独立的激光光路，所述第二成像系统通过第二接收装置获取到第二像的位置的不同判断样品高度的变化，根据样品高度的变化进行自动补偿，以保证样品始终处于聚焦的位置。本发明可通过第二成像系统分别对N条独立的激光光路进行光路调节，以实现N条独立的激光光路具有相同的穿透深度，进而无需在切换不同光源时，再进行光路调节，从而节省了实验当中费时的光路调节。
